هدف: هدف از پژوهش حاضر بررسی رابطه بین پنج صفت بزرگ شخصیت با نشانه‌های افسردگی و نقش میانجی گر عزت نفس و خودکارآمدی در دانشجویان بود. روش: روش پژوهش توصیفی از نوع همبستگی بود. به منظور انتخاب نمونه 400 نفر از دانشجویان مشغول به تحصیل دانشگاه آزاد و دولتی شهرستان گنبدکاووس به روش تصادفی خوشه‌ای انتخاب شدند. ابزارهای مورد استفاده در این پژوهش شامل پرسشنامه‌های افسردگی، پنج عاملی شخصیت، عزت نفس و خودکارآمدی بود. برای تجزیه و تحلیل داده‌ها از آزمون همبستگی پیرسون و روش تحلیل مسیر استفاده شد. یافته‌ها: نتایج حاکی از آن بود که روان آزردگی به عنوان قوی ترین پیش بین افسردگی، اثر مثبت معناداری بر آن داشت. درحالی که برونگرایی، توافق گرایی، عزت نفس، خودکارآمدی اثر منفی معناداری بر افسردگی داشتند. هم‌چنین نتایج تحلیل مسیر حاکی از این بوده که متغیرهای عزت نفس و خودکارآمدی به طور همزمان در پیش‌بینی افسردگی براساس  صفات شخصیت  (برون گرایی، توافق گرایی و روان آزردگی) نقش واسطه را ایفا می‌کنند.نتیجه‌گیری: باتوجه به نتایج پژوهش و نقش واسطه‌ای عزت نفس و خودکارآمدی در ارتباط صفات شخصیت و افسردگی، برنامه‌ریزی جهت ارتقای سطح عزت نفس و خودکارآمدی می‌تواند نقش مهمی در کاهش افسردگی به واسطه تقویت صفات شخصیتی برون گرایی و توافق گرایی و کاهش روان آزردگی داشته باشد.
